Real-Time Monitoring of Antibodies

In-line detection of antibodies and fragments breakthrough during affinity purification.

The analytical methods currently used for detecting product breakthroughs in bioproduction often suffer from limited specificity and sensitivity. Our technology enables real-time, in-line detection of product breakthroughs, transforming the way antibody purification is monitored. This system enables highly sensitive detection of IgG and antibody fragments directly in the effluent stream of affinity chromatography processes.  

Unlike traditional UV methods, which are often saturated or inaccurate due to complex sample backgrounds, the LSPR sensor offers picometer-resolution detection at IgG and antibody concentrations as low as 2.5 µg/mL. The sensor’s functionalized chips provide exceptional selectivity, while in-line regeneration allows for continuous, multi-cycle operation.  

This robust and scalable solution significantly improves breakthrough detection, supports process automation, and paves the way for fully integrated continuous biomanufacturing.

Antibody Aggregate Monitoring

Rapid at-line quantification and in-line detection of antibody aggregates in bioprocessing.

Antibody aggregates are formed when individual antobodies clump together, either during production, storage, or handling of therapeutic antibodies and diagnostic reagents.  

In therapeutic applications, these aggregates may increase immunogenicity, triggering unwanted immune responses in patients. Our cutting-edge nanoplasmonic biosensor technology offers rapid quantification of antibody aggregates and in-line detection of aggregates in affinity purification.  

This technology differentiates monomers from aggregates based on distinct binding kinetics. It offers unparalleled sensitivity, detecting low levels of aggregate, a feature not achievable with traditional UV detection alone. This system supports continuous monitoring, automation, and enhanced quality control. The ability to detect aggregates in real time streamlines downstream purification and strengthens compliance with emerging process analytical technology (PAT) and Quality by Design (QbD) frameworks.

Case Study

In the experiment below, we compared five IgG samples with aggregates ranging from 1 to 20%. In the UV-signal, no variation can be seen for the different samples but with a sensor from ArgusEye it is possible to detect and differentiate samples with aggregate levels.

  • Aggregate levels in the elition pool can be measured in real-time.
  • Lower aggregate levels (<1%) can be determined if higher load is used.
